시설장비 설명 | 독일의 Carl Zeiss사의 Axioskop 2 모델 현미경의로써 사용법은 아래와 같음. I. Brightfield Microscopy Use A. Setting up Koehler Illumination Transmitted-light brightfield microscopy allows for viewing of high-contrast or stained specimens. (Nonfluorescent specimens e.g. H&E slides alkaline phosphatase localization). To utilize the entire optical performance of the microscope especially of the objective the condenser the luminous-field diaphragm and the aperture diaphragm should be set in accordance with the rules for KOEHLER illumination. TO SET UP KOEHLER ILLUMINATION: 1. Turn on halogen lamp with ON/OFF switch on back of microscope stand (3-1/27). The green pilot lamp (3-1/10) will light up. 2. Place a high-contrast specimen (e.g. H&E slide) on the mechanical stage (3-1/21) the coverslip pointing upwards. 3. Swing in 10X objective on the nosepiece and focus on the specimen. NOTE: The front lens of the condenser must be swung IN (using lever 3-2/2) for all objectives above 5X. 4. Make sure the reflector turret (3-1/23) is turned to position 4 or 5 (empty for brightfield viewing). 5. Set image brightness via voltage (light intensity) control (3-1/11). 6. Move condenser (3-1/20) to the upper stop position via the condenser drive (3-1/25). 7. Close luminous-field diaphragm (3-1/24) until it is visible in the field of view (Fig. 01/A). 8. Lower condenser until the edge of the luminous-field diaphragm appears in focus (Fig. 01/B). 9. Center luminous-field diaphragm using centering screws (3-1/19) on the condenser carrier (Fig. 01/C) and open it until its edge just disappears behind the field of view (Fig. 01/D). 10. To set the aperture diaphragm (contrast) remove one eyepiece (3-1/1) from the binocular tube and using the sliding knob on the front of the condenser (3-2/3) set the aperture diaphragm to approximately 2/3 to 4/5 of the diameter of the objective exit pupils (Fig. 01/E). 11.
